Genomic Instability: The accumulation of DNA damage and mutations over time, combined with the age-related decline in DNA repair mechanisms, is a central driver of both aging and cancer. Age-associated changes in gene expression patterns (without altering the DNA sequence itself), such as DNA methylation shifts and chromatin remodeling, can silence tumor suppressor genes or activate oncogenes, thereby contributing to cancer initiation and progression. Aging is associated with a low-grade, chronic state of inflammation. This condition creates a tumor-promoting microenvironment through the continuous release of pro-inflammatory cytokines and growth factors.
